Preparation of RNA samples for sequencing on the Illumina HiSeq. Described here is a library preparation service only. Once the library is prepared, you will also need to request (and pay for) the sequencing of that library. Total RNA is input into the workflow where mRNA is isolated using oligo d(T), cDNA is made with reverse transcriptase. The cDNA is end-repaired A tailed and Illumina adapters are ligated. The resultant library is then PCR amplified to have enough material for the sequencer.

Sample Requirements10-500ng of ChIP enriched DNA, up to 550bp
Critical Information
  1. input DNA is the appropriate comparison to ChIPped DNA, not an IgG control pulldown;

  2. a reference NON-pulldown gene is essential; and

  3. we have doubts about samples that don't show a 10-fold enrichment by qPCR performed as a delta-delta Ct
Additional Information
  1. Due to limited space in the cores freezer's we can only keep samples for 6 months. If you would like your samples returned to you after sequencing please contact the core.

  2. Due to size limitations on the HiSeq 4000, libraries with a average bp great than 580bp can not be run on the HiSeq 4000.
